Adipose triglyceride lipase regulates eicosanoid production in activated human mast cells

Human mast cells (MCs) contain TG-rich cytoplasmic lipid droplets (LDs) with high arachidonic acid (AA) content. Here, we investigated the functional role of adipose TG lipase (ATGL) in TG hydrolysis and the ensuing release of AA as substrate for eicosanoid generation by activated human primary MCs in culture. Silencing of ATGL in MCs by siRNAs induced the accumulation of neutral lipids in LDs. IgE-dependent activation of MCs triggered the secretion of the two major eicosanoids, prostaglandin D2 (PGD2) and leukotriene C4 (LTC4). The immediate release of PGD2 from the activated MCs was solely dependent on cyclooxygenase (COX) 1, while during the delayed phase of lipid mediator production, the inducible COX-2 also contributed to its release. Importantly, when ATGL-silenced MCs were activated, the secretion of both PGD2 and LTC4 was significantly reduced. Interestingly, the inhibitory effect on the release of LTC4 was even more pronounced in ATGL-silenced MCs than in cytosolic phospholipase A₂-silenced MCs. These data show that ATGL hydrolyzes AA-containing TGs present in human MC LDs and define ATGL as a novel regulator of the substrate availability of AA for eicosanoid generation upon MC activation.     

Localization of a gene for a minor chorion protein in Drosophila melanogaster: a new chorion structural locus

A minor chorion protein (called s70) with an approximate molecular weight of 70,000 D has been characterized in Drosophila melanogaster. The Staket geographic strain was found to carry an electrophoretic variant of this eggshell component and was used to determine the chromosomal location of the s70 gene. Our results establish a new locus for a chorion gene near yellow on the X chromosome and represent the first mapping of a quantitatively minor eggshell protein.     

Crosslinking of theDrosophila chorion involves a peroxidase

Summary TheDrosophila chorion contains an endogenous peroxidase activity which remains inactive until late stage 14 when it catalyzes the crosslinking of the chorionic proteins. Using explanted follicles developing in vitro, premature, but otherwise normal crosslinking can be induced with hydrogen peroxide and normal crosslinking can be prevented with peroxidase inhibitors. Inhibition or premature activation of the shell peroxidase allows characterization of chorionic filament specific proteins and establishes new criteria for the identification of eggshell components.     

Predation risk and feeding patterns of crucian carp

As part of an experimental study of the direct and indirect effects of piscivory on prey fish, the diets of crucian carp Carassius carassius were compared across sections of a divided pond; two sections were stocked with crucian carp alone and two with crucian carp plus perch Perca fluviatilis . Analysis of crucian gut contents indicated that the composition of invertebrate prey did not differ in the presence v . absence of perch. However, crucians, particularly small individuals (<10cm) that were most vulnerable to predation, displayed a lower intake of invertebrate prey in sections with perch. Although diet composition differed between crucians caught in inshore v. offshore habitats (with habitat use being related to crucian size and the presence or absence of perch), no clear pattern existed between habitat and total food intake. Overall, the major effects of predators on the diet of crucian carp appeared to be caused by increased ecological density (resulting from confinement of small crucians inshore) and reduced activity levels, rather than simple shifts to safer habitats.

Both in the stems woumded let free and in those in which an extra tissue has been introduced, the cicatrization processes tacke place. They only take place with different velocity. The healing process lead to the reconstruction of the cortical and the conducting tissues.

The whole reconstruction of the stele may occur, when the flow of the assimilated stuffs is slowed. Sothat it is probable that the regeneration depends more upon the tissue nutrition tham on a specifical properti of the tissues.     

The production of specific antisera to bradykinin and their study by immunoenzyme analysis

Bradykinin coupled to bovine serum albumin or to rabbits erythrocytes was used to immunize rabbits. It was shown that enzyme-linked immunosorbent assay (ELISA) has been successfully used for the investigation of the obtained immune sera.
